However, Glasgow can take confidence from a 12-6 home victory over Leinster last September, one of just four regular-season defeats for the champions.
Munster, who suffered an agonising 16-15 loss to Glasgow in the semi-finals, host Edinburgh in the opening round.
Ulster also reached the play-offs last season and travel to the Scarlets for their first fixture.
Other season-opening clashes, set to take place between September 5 and 7, see Newport Gwent Dragons visit Connacht, Ospreys host Treviso and Zebre take on Cardiff Blues in Parma.
