Hagino faced a late challenge from the aptly named Chase Kalisz, but the American was made to settle for silver as Hagino touched the wall in four minutes and 6.05 seconds.
The event kicked off an evening programme that will see a total of four gold medals handed out at the Rio de Janeiro's Aquatics Stadium.
