Having beaten Japan's Misato Nakamura in the semi-final to guarantee at least silver, two-time world champion Kelmendi triumphed against Odette Giuffrida of Italy in the gold medal match.
Kelmendi competed for Albania at the 2012 Games in London and coincidentally won her first world title in Rio in 2013, with a single yuko enough to seal her historic victory against Giuffrida on this occasion.
Kosovo, which declared independence from Serbia in 2008, is participating in the Olympic Games for the first time after being granted the right to compete by the International Olympic Committee two years ago.
