Troy Brouwer was the hero for Washington, scoring two minutes and 12 seconds into overtime as the Capitals edged the Bruins 5-4 at Verizon Center.
The Capitals, who narrowly missed out on a wildcard spot last season, trailed 4-2 but fought back as Michael Latta and Liam O'Brien found the net.
Alex Ovechkin had two power-play goals for the Capitals, while Washington goaltenders Philipp Grubauer and Justin Peters made 19 saves between them.
Seth Griffin led the way for the Bruins, who reached the conference semi-finals in 2013-14.
Griffin scored twice, while Matt Fraser and Matt Lindblad had one apiece.
Bruins goaltender Niklas Svedberg made 24 saves.
Last season's runners-up, the Rangers were also triumphant after they downed the Chicago Blackhawks 4-1.
Goals from Oscar Lindberg, Anthony Duclair, JT Miller and Marek Hrivik inspired the Rangers to a routine win at United Center.
In other results, Kyle Okposo and Brock Nelson all scored in the shoot-out as the New York Islanders prevailed 3-2 against the New Jersey Devils.
Anton Stralman scored twice as the Tampa Bay Lightning accounted for the Dallas Stars 6-3.

Henrik Sedin assisted on three power-play goals as the Vancouver Canucks shutout the Calgary Flames for a 3-0 win.
